Proteins, carbohydrates and fats are natural macromolecules. The partial structure of a protein is shown below. [Figure shows a repeating peptide linkage structure: H O H H O H H O H H | || | | || | | || | | || | N–C–C–N–C–C–N–C–C–N–C–C–N– C C C C C | | O O O O ]

  1. (a)Name the linkage that joins the monomer units in a protein. [1][1]
  2. (b)Name a synthetic polymer that has the same linkage as a protein. [1][1]
  3. (c)Proteins are hydrolysed to give a mixture of colourless amino acids. Describe, with the aid of a labelled diagram, how paper chromatography can be used to identify the amino acids present in a mixture of amino acids. .[4][4]
  4. (d)Carbohydrates can be hydrolysed. Name the class of compound formed when carbohydrates are hydrolysed. [1][1]
  5. (e)(i)The diagram shows the structure of a simple fat. [Figure shows a triglyceride structure: H | H-C-O-C-C15H27 || O | H-C-O-C-C15H27 || O | H-C-O-C-C15H27 || H O ] This fat is polyunsaturated. What is the meaning of the term polyunsaturated? .[2][2]
  6. (e)(ii)Describe a chemical test to show that the fat is unsaturated. name of reagent result of test ..[2][2]
  7. (e)(iii)Name a synthetic macromolecule that contains the same linkage as fats. .[1][1]

Mark scheme

Marking points from the official mark scheme (May/June 2014). The mark or mark type is shown in brackets.

  1. [1] A3 (a) Amide / peptide (1)
  2. [1] (b) Nylon / Kevlar (1) ALLOW: polyamide
  3. [1] (c) Chromatography paper dipped into the solvent (1) ALLOW: chromatography paper just touching solvent (there should be no space between the solvent and the bottom of the paper)
  4. [1] (c) Spot of mixture on paper above the level of the solvent labelled appropriately e.g. mixture / amino acid / amino acids / spot of amino acid / drop from sample (1)
  5. [1] (c) Use of a locating agent to view the spots / amino acids / use of ninhydrin to view spots / amino acids (1) NOTE: this must be after the chromatography
  6. [1] (c) Comparing Rf values with known amino acids / compare with height of spots from known amino acids run at the same time (1)
  7. [1] (d) (simple) sugars / monosaccharides (1)
  8. [2] (e) (i) Has many C=C bonds / has many carbon-carbon double bonds (2) If two marks not scored, award 1 mark for has C=C double bond.
  9. [1] (e) (ii) Bromine / bromine water (1)
  10. [1] (e) (ii) Decolourised / goes colourless (1) IGNORE: goes clear / discoloured / fades NOTE: second mark dependent on correct reagent
  11. [1] (e) (iii) Polyester / Terylene / other named polyester (1)
